Oxytocin is a cyclic nonapeptide (CYIQNCPLG-NH2) synthesised in the paraventricular nucleus (PVN) and supraoptic nucleus (SON) of the hypothalamus and released from posterior pituitary terminals. It acts via the oxytocin receptor (OXTR), a Gq/11-coupled GPCR, activating phospholipase C-β and downstream IP3/DAG/PKC cascades. Research models encompass social cognition, fear attenuation, HPA axis regulation, uterine contractility, and lactation — making it one of the most studied neuroendocrine peptides in behavioural neuroscience.

Mechanism of Action

Oxytocin's actions are mediated by a single receptor type, OXTR (oxytocin receptor), which exhibits high structural homology to the V1a vasopressin receptor (they diverged from a common ancestral neuropeptide receptor). OXTR couples primarily to Gq/11, activating phospholipase C-β (PLCβ), which hydrolyses PIP2 to generate IP3 and DAG. IP3 triggers intracellular Ca2+ release from the endoplasmic reticulum, while DAG activates protein kinase C (PKC). This Ca2+/PKC signalling cascade mediates uterine and mammary myoepithelial contraction in peripheral tissues. OXTR also signals via Gi in some cell types, reducing cAMP, and activates ERK1/2 and Akt through β-arrestin-biased pathways.

In the central nervous system, oxytocinergic projections from PVN neurons reach the amygdala, nucleus accumbens (NAc), prefrontal cortex, hippocampus, and brainstem. Within the amygdala, OXTR activation on GABAergic interneurons increases inhibitory tone on CRH-expressing neurons of the central nucleus (CeA), reducing fear responses and stress reactivity. In the NAc, oxytocin potentiates dopaminergic neurotransmission at D1 receptors, enhancing social reward salience. This mesolimbic gating underlies the extensive research into oxytocin's role in affiliative behaviour and social memory consolidation.

Regarding HPA axis modulation, oxytocin neurons in the PVN co-release oxytocin with CRH from the same parvocellular population in some species; however, OXT predominantly inhibits HPA activation by suppressing CRH neuron firing via autocrine OXTR signalling and by direct inhibition of ACTH secretion from anterior pituitary corticotrophs. This provides an endogenous anxiolytic/anti-stress circuit of significant interest in stress neuroscience research.
